Vitex agnus-castus, commonly known as chaste tree berry, has a long history of traditional use in supporting female reproductive health, and modern scientific research has begun to elucidate its effects on the endocrine system, particularly through actions involving the hypothalamus-pituitary axis. Several studies indicate that Vitex agnus-castus may influence the secretion of prolactin by acting on dopamine receptors in the hypothalamus, which in turn affects pituitary hormone release. This mechanism underlies its use in conditions such as premenstrual syndrome (PMS), cyclic mastalgia, and certain menstrual irregularities. Clinical trials have shown that supplementation with Vitex can reduce symptoms of PMS, likely via modulation of prolactin and possibly through indirect effects on luteinizing hormone (LH) and follicle-stimulating hormone (FSH) levels, both of which are regulated by the hypothalamus.
While the precise mechanisms remain an active area of research, systematic reviews and randomized controlled trials support the notion that Vitex agnus-castus exerts neuroendocrine effects that can be traced back, at least in part, to hypothalamic modulation. However, evidence is not yet robust for all hypothalamic-associated conditions, and more research is needed to clarify its broader role. Overall, the evidence for Vitex’s support of the hypothalamic-pituitary axis, particularly in the context of female hormonal balance, is moderate and based on both mechanistic and clinical studies.
