Several clinical trials have investigated undenatured type II collagen (UC-II) for the management of joint discomfort, particularly in individuals with osteoarthritis and rheumatoid arthritis.
Some studies have shown that supplementation with UC-II can help improve joint function and reduce joint pain and stiffness compared to placebo. For example, a randomized, double-blind, placebo-controlled study published in International Journal of Medical Sciences (2013) demonstrated that daily supplementation with UC-II improved knee joint symptoms in osteoarthritis patients. The proposed mechanism involves oral tolerance, where small amounts of collagen II help modulate immune responses that target joint cartilage.
However, the quality and size of studies are variable, and some reviews note that while results are promising, more large-scale, high-quality trials are needed to establish conclusive efficacy. Collagen II is also sometimes compared to glucosamine and chondroitin, other popular joint supplements, with some studies suggesting similar or superior benefits.
