N-acetylglucosamine (NAG) is a derivative of glucose and a component of glycosaminoglycans, which are important for the structure of joint cartilage. Its use for joint support is based on the premise that oral supplementation may contribute to the repair or maintenance of cartilage and synovial fluid. There is some scientific rationale for its use, as NAG is a building block of hyaluronic acid and other molecules present in joint tissues. However, the majority of clinical research focuses on glucosamine sulfate or glucosamine hydrochloride, rather than NAG specifically.

A limited number of studies have investigated NAG for joint health directly. Some small-scale clinical trials and animal studies suggest potential benefits for joint-related symptoms, such as reduced pain and improved function, especially in osteoarthritis. However, these studies are generally few, of limited size, and sometimes of low methodological quality.
