Acemannan is a polysaccharide extracted from the inner leaf gel of Aloe vera, and its use for gastrointestinal issues, including Leaky Gut Syndrome (increased intestinal permeability), is supported by preliminary scientific evidence rather than a long-standing traditional use. Several in vitro and animal studies have shown that acemannan can promote intestinal epithelial cell proliferation, enhance mucosal healing, and modulate immune responses. For example, studies in rodents have demonstrated that acemannan can help restore tight junction integrity in the intestinal lining and reduce inflammation associated with gut barrier dysfunction. However, there is a lack of robust human clinical trials specifically evaluating acemannan for Leaky Gut Syndrome. Most evidence comes from laboratory studies or research in related contexts, such as inflammatory bowel disease models. While these findings are promising and suggest a potential role for acemannan in supporting gut barrier function, the evidence is not considered strong (hence a rating of 2/5). More rigorous clinical trials are needed to confirm its efficacy and safety for this specific indication in humans.
