Glutamine is an amino acid that plays a key role in intestinal health and is commonly used to support "leaky gut syndrome," a term used to describe increased intestinal permeability. While "leaky gut syndrome" is not universally recognized as a formal medical diagnosis, increased intestinal permeability is a documented phenomenon in certain diseases. Scientific studies have shown that glutamine is a major fuel source for enterocytes (cells lining the intestine) and can help maintain the integrity of the gut barrier. Some animal and in vitro studies suggest that glutamine supplementation can reduce intestinal permeability and improve mucosal healing, particularly in stressed or injured gut tissues. Small clinical studies have explored glutamine’s effects in critically ill patients, those undergoing chemotherapy, or patients with inflammatory bowel disease, and some have shown benefits in reducing markers of gut permeability. However, high-quality, large-scale clinical trials directly evaluating glutamine’s effectiveness for "leaky gut syndrome" in otherwise healthy individuals are lacking. Thus, while there is a plausible scientific rationale and some preliminary supporting data, the overall evidence in humans for this specific application is limited, justifying a moderate evidence rating.
