Mannanoligosaccharides (MOS) are prebiotic fibers derived primarily from the cell walls of yeast (Saccharomyces cerevisiae). Their purported benefit for “leaky gut syndrome”—a non-medical term often used to describe increased intestinal permeability—arises from their ability to modulate gut microbiota and enhance gut barrier function. Preclinical studies, mostly in animal models, suggest that MOS can reduce intestinal permeability, modulate immune responses, and decrease pathogen adhesion to the gut wall. For example, studies in piglets and poultry have demonstrated that dietary MOS administration improves gut integrity and reduces markers of inflammation and permeability. The proposed mechanisms involve modulation of the gut microbiome and stimulation of the immune system, possibly leading to improved tight junction integrity in the intestinal lining.

However, direct human clinical evidence for MOS specifically improving intestinal permeability or treating "leaky gut syndrome" is limited. Most available data are from animal studies or extrapolated from related prebiotics. While some human studies suggest benefits of prebiotics in general for gut health, conclusive evidence for MOS in humans for this condition is lacking. Thus, while there is some scientific basis for its use (mostly preclinical), the evidence is preliminary rather than robust, justifying a moderate rating.
