Transforming growth factor-beta (TGF-β) is a cytokine known to play a crucial role in tissue repair and regeneration, including the healing of ligaments. Scientific research demonstrates that TGF-β is actively involved in the regulation of cellular processes critical to ligament healing, such as cell proliferation, differentiation, and extracellular matrix production. Animal studies and in vitro experiments consistently show that supplementation or increased expression of TGF-β enhances collagen synthesis and deposition, accelerates the proliferation of fibroblasts, and improves the structural organization of healing ligament tissue.

For example, studies in rat models of medial collateral ligament (MCL) injury reveal that local application of TGF-β significantly improves the mechanical strength and histological architecture of the repaired ligament. Additionally, research in rabbits and other animal models supports the hypothesis that TGF-β can accelerate the early phase of ligament healing and reduce scar formation. These findings have informed the development of tissue engineering approaches, where TGF-β is incorporated into scaffolds or delivery systems to enhance ligament regeneration.

However, while preclinical evidence is robust, clinical trials in humans remain limited. Early-phase human studies and case reports suggest potential benefits but are not yet definitive. Overall, there is substantial scientific validation in preclinical models for the use of TGF-β in treating ligament injuries, supporting further research and development in this area.
