D-glucarate, particularly in the form of calcium D-glucarate, is commonly marketed as a supplement to support liver detoxification. The scientific rationale behind its use centers on its ability to inhibit the enzyme beta-glucuronidase. Beta-glucuronidase is involved in the deconjugation of toxins and hormones (such as estrogens) that have been made water-soluble in the liver for excretion. Elevated activity of this enzyme in the gut can lead to the reactivation and reabsorption of these potentially harmful compounds, which may increase the body's toxic burden.

Animal studies and in vitro research have demonstrated that D-glucarate and its metabolites can decrease beta-glucuronidase activity and increase the excretion of carcinogens, estrogens, and other toxins. Some rodent studies suggest that calcium D-glucarate supplementation may inhibit chemical-induced cancers, particularly those of the liver, breast, and colon, through this mechanism. However, there is a lack of robust clinical trials in humans directly assessing D-glucarate's effect on liver detoxification or health outcomes. Most evidence is preclinical or observational, and the translation of these findings to human detoxification processes remains uncertain.

In summary, while there is a scientific basis for the proposed mechanism of D-glucarate in supporting phase II liver detoxification (glucuronidation), high-quality clinical evidence in humans is limited. Its use is justified by mechanistic and animal data, but direct proof of benefit in human liver detoxification is not yet established.
