Dithiolthiones, such as 1,2-dithiole-3-thione (D3T) and oltipraz, are sulfur-containing compounds that have been investigated for their potential to support liver detoxification. Their use is primarily justified by scientific research rather than tradition. Several preclinical and limited clinical studies have shown that dithiolthiones can induce phase II detoxification enzymes, especially glutathione S-transferases (GSTs), in the liver. These enzymes play a crucial role in conjugating and eliminating toxins and carcinogens.
Oltipraz, for example, has been studied as a chemopreventive agent because it increases the expression of detoxification enzymes and protects against chemically induced liver damage in animal models. Some early phase clinical trials in humans have suggested that oltipraz can modulate these detoxification pathways, but robust clinical evidence for its efficacy in general liver "detoxification" or in treating liver disease is lacking. The evidence supporting the use of dithiolthiones for liver detoxification is primarily mechanistic and preclinical, with moderate support from small human trials focusing on specific toxin exposures rather than general health claims.
In summary, dithiolthiones have a scientific rationale for supporting liver detoxification processes at the biochemical level, but the clinical evidence for broad use in liver detoxification or disease treatment remains limited. More large-scale, controlled human studies are needed to fully validate their efficacy and safety for this purpose.
