Interleukins are a group of cytokines (signaling molecules) that play a key role in the immune system. In the context of lymphoma, certain interleukins have been investigated and used therapeutically, most notably interleukin-2 (IL-2). IL-2 is an FDA-approved biologic agent that stimulates the growth and activity of T cells and natural killer (NK) cells, which can help attack cancer cells, including in some lymphomas. Clinical studies have evaluated IL-2, often in combination with other treatments, in various types of lymphoma such as cutaneous T-cell lymphoma (CTCL) and non-Hodgkin lymphoma (NHL). Results have been mixed: while some patients experience tumor regression or disease stabilization, response rates are generally modest, and IL-2 is associated with significant toxicities that limit its widespread use. Other interleukins, like IL-12 and IL-15, are under investigation, but are not yet standard therapies. The use of interleukins is supported by mechanistic rationale and clinical trial data, but they are not considered first-line or curative treatments for lymphoma. Their use is reserved for select cases, often within clinical trials. Thus, there is scientific evidence for the use of certain interleukins in lymphoma, but the strength of evidence and clinical benefit is moderate and continues to be an area of active research.
