Creatine phosphate (also known as phosphocreatine) is best known for its role in energy metabolism in muscle cells, but there is emerging scientific interest in its effects on the brain. Creatine plays a critical role in cellular energy homeostasis, and the brain—being a highly energy-dependent organ—can potentially benefit from enhanced creatine availability. Some small-scale clinical studies and animal experiments suggest that creatine supplementation may have mild cognitive benefits, particularly in situations of metabolic or energetic stress (e.g., sleep deprivation, mental fatigue, or in elderly populations).
Randomized controlled trials have shown modest improvements in tasks involving short-term memory and reasoning in healthy young adults and vegetarians (who have lower baseline creatine stores). There is also preliminary evidence that creatine supplementation may help memory performance and reduce mental fatigue in the elderly and in neurological conditions, such as traumatic brain injury. However, the number of high-quality studies is still limited, and the effects observed are generally small. The mechanisms are hypothesized to involve enhanced ATP availability and neuroprotection under stress, but these have not been definitively established in humans.
