β-nicotinamide mononucleotide (NMN) is a precursor to nicotinamide adenine dinucleotide (NAD+), an essential coenzyme involved in cellular energy metabolism and redox reactions. Scientific interest in NMN’s potential to support or treat Metabolic Syndrome stems from preclinical studies demonstrating that NAD+ levels decline with age and metabolic dysfunction, and that restoring NAD+ via NMN supplementation may improve metabolic health. In animal models, NMN administration has been shown to enhance insulin sensitivity, reduce hepatic fat accumulation, improve lipid profiles, and increase energy expenditure, all key factors in Metabolic Syndrome. Early-phase human studies indicate that NMN is generally safe and may modestly improve insulin sensitivity in overweight or prediabetic individuals; for example, a 2021 randomized controlled trial in postmenopausal women with prediabetes reported improved muscle insulin sensitivity after NMN supplementation. However, these human trials are limited in number, size, and duration, and results are not uniformly positive. There is currently insufficient robust clinical evidence to conclude that NMN supplementation is an effective treatment for Metabolic Syndrome in humans, but the biological rationale and promising preclinical findings have spurred ongoing research. NMN is not rooted in traditional medicine but rather in recent advances in molecular biology and metabolic research. Thus, while scientific validation is emerging, it remains preliminary, and NMN should not yet be considered a proven therapy for Metabolic Syndrome.
