Nicotinamide Adenine Dinucleotide (NAD+) is a critical coenzyme involved in cellular metabolism, energy production, and redox reactions. Its role in metabolic health has garnered scientific interest, particularly regarding conditions like metabolic syndrome—a cluster of risk factors including obesity, insulin resistance, hypertension, and dyslipidemia. Experimental and early clinical studies indicate that NAD+ levels decline with age and metabolic stress, impairing mitochondrial function and contributing to metabolic dysfunctions. Supplementation with NAD+ precursors—such as nicotinamide riboside (NR) or nicotinamide mononucleotide (NMN)—has been shown in animal models to improve insulin sensitivity, reduce inflammation, and enhance mitochondrial function.

Human data are more limited but growing. Small-scale clinical trials have suggested that NAD+ precursor supplementation can modestly improve certain metabolic parameters (e.g., insulin sensitivity, lipid profiles) in overweight or prediabetic individuals.
