Specialized pro-resolving mediators (SPMs), including resolvins, protectins, and maresins, are bioactive lipid compounds derived from omega-3 fatty acids and are known for their roles in actively resolving inflammation. There is an emerging scientific interest in the potential of SPMs to support migraine management, given the established connection between neuroinflammation and migraine pathophysiology. Preclinical studies suggest that SPMs may reduce pain signaling and neuroinflammation by modulating immune cell activity and supporting the resolution of inflammatory processes in the nervous system. Some animal models of migraine have demonstrated that administration of SPMs can reduce migraine-like pain behaviors and inflammatory markers.

However, human clinical evidence supporting the effectiveness of SPMs in treating or preventing migraines is currently extremely limited. No large-scale, well-controlled clinical trials have been published to date. Most available data are derived from basic research or small pilot studies, and the precise mechanisms by which SPMs might influence migraine frequency or severity in humans remain unclear. As such, while the theoretical rationale for SPM use in migraine exists and some preliminary animal data are promising, the overall quality and quantity of scientific evidence is low at this time. More rigorous human studies are needed before SPMs can be recommended as a validated treatment or support for migraine.
