Calcium alpha-ketoglutarate (Ca-AKG) is a salt form of alpha-ketoglutarate, a key intermediate in the Krebs (citric acid) cycle, which is central to mitochondrial energy production in cells. Its use as a supplement to support mitochondrial function is based on its biochemical role within this cycle. Animal studies and in vitro experiments have shown that alpha-ketoglutarate can enhance cellular energy metabolism, reduce oxidative stress, and promote mitochondrial health. For example, several rodent studies have demonstrated improved mitochondrial function and extended lifespan with AKG supplementation, possibly due to effects on cellular metabolism and modulation of age-related pathways. In addition, some research suggests AKG may reduce accumulation of certain metabolic byproducts that impair mitochondrial efficiency.

However, direct evidence in humans remains limited. The majority of studies have been conducted in animal models, and there is a lack of large, high-quality human trials demonstrating clinical benefits for mitochondrial health or related outcomes. While the scientific rationale for supplementation is reasonable given AKG’s central role in cellular energy production, the current level of evidence supporting Ca-AKG specifically for mitochondrial support in humans is modest. Thus, the rating reflects promising but preliminary scientific validation, with further research needed to confirm efficacy and safety in humans.
