Cardarine (GW501516) is a peroxisome proliferator-activated receptor delta (PPAR-δ) agonist, initially developed for its potential to treat metabolic and cardiovascular diseases. Its relevance to mitochondrial function comes from its ability to enhance the expression of genes involved in fatty acid oxidation and energy metabolism, particularly in muscle tissue. Preclinical studies in rodents have shown that Cardarine increases mitochondrial biogenesis and improves mitochondrial efficiency, leading to improved exercise endurance and lipid metabolism (Narkar et al., 2008). These effects are mediated by upregulation of pathways that enhance oxidative metabolism, mimicking some aspects of endurance exercise at the cellular level. However, most of the evidence for these mitochondrial benefits comes from animal studies and cell culture models. Few, if any, robust clinical trials in humans have confirmed these effects or established their safety profile. Cardarine’s development for clinical use was halted due to concerns about carcinogenicity observed in long-term animal trials. Nevertheless, its mitochondrial effects are scientifically plausible and supported by mechanistic data, though human relevance and safety remain unproven. Therefore, while there is scientific basis for Cardarine’s support of mitochondrial function, the evidence is incomplete and not clinically validated.
