Japanese knotweed (Reynoutria japonica, formerly Polygonum cuspidatum) is a plant rich in polyphenols, especially resveratrol. Resveratrol has been studied for its potential to support mitochondrial health through its effects on cellular energy metabolism and antioxidant defenses. In laboratory and animal studies, resveratrol has been shown to activate sirtuins (notably SIRT1), which can improve mitochondrial biogenesis and function. These actions are thought to be mediated in part by activation of the PGC-1α signaling pathway, a master regulator of mitochondrial biogenesis (Lagouge et al., 2006). Additionally, resveratrol’s antioxidant properties may help protect mitochondria from oxidative damage. However, most of the evidence comes from in vitro and animal research, with limited direct clinical evidence in humans regarding knotweed or its extracts specifically supporting mitochondrial function. Human trials with resveratrol (from various sources) show mixed results, and optimal dosing or clinical benefits remain unclear. Knotweed itself has a long history of use in traditional medicine, but its application for mitochondrial support is based primarily on its resveratrol content and modern biochemical research rather than established traditional use. Therefore, while knotweed contains compounds with plausible mechanisms for mitochondrial support, the evidence in humans is still preliminary.
