Magnesium Alpha-Ketoglutarate (Mg-AKG) is a compound combining magnesium, an essential mineral, and alpha-ketoglutarate (AKG), a key intermediate in the Krebs (citric acid) cycle within mitochondria. The rationale for its use in supporting mitochondrial function is grounded in the biological roles of both components. Magnesium is well-established as a cofactor in hundreds of enzymatic reactions, many of which occur in mitochondrial energy metabolism. AKG, on the other hand, is directly involved in the Krebs cycle, which is central to cellular ATP production. Some animal and in vitro studies have shown that AKG supplementation can enhance mitochondrial function, reduce oxidative stress, and promote cellular energy production. However, direct clinical evidence in humans using Mg-AKG specifically for mitochondrial support is limited. Most human studies focus on AKG or magnesium individually, and while both show benefit in supporting general cellular metabolism and reducing fatigue, robust clinical trials specifically investigating Mg-AKG for mitochondrial health are lacking. In summary, the use of Magnesium Alpha-Ketoglutarate to support mitochondrial function is scientifically plausible based on its biochemical roles, and there is some preclinical evidence.
