NADH (nicotinamide adenine dinucleotide + hydrogen) is a coenzyme found in all living cells and plays a critical role in mitochondrial energy production. In the mitochondria, NADH acts as an electron donor in the electron transport chain, helping generate ATP—the cell’s main energy currency. This biochemical function is well established in scientific literature and is universally recognized in cellular biology.

Supplemental NADH has been studied for its potential to enhance mitochondrial function and energy metabolism, particularly in conditions characterized by fatigue such as chronic fatigue syndrome (CFS) and fibromyalgia. Some small clinical trials and pilot studies suggest that NADH supplementation may improve energy levels and cognitive function in such populations, but results are mixed and not consistently reproducible. There’s also limited evidence for its use in neurodegenerative diseases like Parkinson’s disease, with some early studies noting modest benefits, but again, data are not robust.

Overall, while the role of NADH within mitochondria is scientifically validated, the evidence for oral NADH supplementation directly supporting mitochondrial health or function in otherwise healthy individuals is limited and not conclusive. Most clinical studies are small, short-term, and often lack rigorous controls. Therefore, while NADH’s use is grounded in its biological role, strong clinical validation for supplementation is lacking, justifying a moderate evidence score.
