Oxaloacetate is a key metabolic intermediate in the citric acid (Krebs) cycle, which is central to mitochondrial energy production. Scientific interest in oxaloacetate supplementation has grown due to its role in cellular metabolism and energy generation. Preclinical research, particularly in animal models, indicates that oxaloacetate supplementation can enhance mitochondrial function, increase the NAD+/NADH ratio, and potentially improve cellular energy status. Some studies have shown benefits in models of neurodegeneration and aging, likely due to improved mitochondrial resilience and bioenergetics. Human research is limited, but a few small pilot studies suggest potential cognitive and energy benefits, though these findings are preliminary and not yet conclusive. Mechanistically, oxaloacetate supports mitochondrial health by facilitating the efficient flow of the Krebs cycle, thereby optimizing ATP production. Additionally, its role in gluconeogenesis and as an antioxidant may further support cellular health under stress. However, there is not yet robust clinical evidence in humans to fully validate its effectiveness for mitochondrial support as a dietary supplement. Most claims are based on mechanistic plausibility and early-stage studies. Thus, while there is a scientific foundation for its use and moderate evidence from animal and pilot human studies, larger and more rigorous clinical trials are needed to establish its efficacy in supporting mitochondrial function in humans.
