Synopsis

Source of validity: Scientific
Rating (out of 5): 3

Phosphatidylethanolamine (PE) is a major phospholipid component of biological membranes, including those of the mitochondria. It plays a critical role in maintaining mitochondrial membrane integrity, shape, and function. Scientific research demonstrates that PE is essential for mitochondrial bioenergetics, as it supports membrane curvature and the assembly of mitochondrial protein complexes involved in oxidative phosphorylation. Mice and yeast models with genetically impaired PE biosynthesis exhibit mitochondrial dysfunction, altered energy metabolism, and increased oxidative stress, indicating that adequate PE is crucial for optimal mitochondrial activity.

Supplementation or increased biosynthesis of PE has been studied in the context of mitochondrial diseases and aging, with some preclinical models showing improved mitochondrial morphology and function. However, direct clinical trials in humans using oral PE supplementation specifically to enhance mitochondrial function are lacking. Most evidence comes from in vitro, animal, and mechanistic studies. PE is sometimes included in mitochondrial support supplements based on its biological role, but the translation from basic science research to clinical benefit in humans is not yet fully established.

In summary, there is scientific rationale and experimental evidence supporting the importance of phosphatidylethanolamine for mitochondrial health, but robust human clinical evidence for supplementation benefits remains limited. Therefore, the evidence rating is moderate.
