Thiamin (vitamin B1) is scientifically validated as essential for mitochondrial function. Thiamin acts as a coenzyme for several key enzymes in cellular energy metabolism, particularly within the mitochondria. It is required for the activity of pyruvate dehydrogenase and alpha-ketoglutarate dehydrogenase, both of which are critical enzymes in the citric acid (Krebs) cycle—a central pathway for mitochondrial energy (ATP) production. Thiamin deficiency impairs these enzymes, leading to reduced ATP synthesis and the accumulation of metabolic intermediates, which can result in symptoms such as fatigue, muscle weakness, and neurological dysfunction.

Multiple studies and reviews have documented that thiamin supplementation can restore mitochondrial function in cases of deficiency, and its use is well-established for preventing and treating conditions like beriberi and Wernicke-Korsakoff syndrome, both of which are associated with mitochondrial dysfunction due to impaired energy metabolism. However, while the requirement for thiamin in mitochondrial energy production is indisputable, evidence for benefit from supplementation beyond correcting deficiency in the general population is limited.
