Triacetyluridine (TAU) is a prodrug of uridine, which is a nucleoside involved in various cellular processes, including mitochondrial function. TAU is more bioavailable than uridine itself, making it effective at increasing systemic uridine levels. Scientific studies, particularly in animal models and some early human research, suggest that uridine supplementation can support mitochondrial function by enhancing mitochondrial RNA synthesis and phospholipid biosynthesis, both crucial for mitochondrial membrane integrity and function. Uridine has also shown potential in improving mitochondrial bioenergetics and protecting against mitochondrial dysfunction, which is implicated in several neurodegenerative and metabolic disorders.

However, while there is a plausible mechanistic basis and some supportive preclinical data, direct clinical evidence for TAU's efficacy in supporting mitochondrial health in humans is limited. Most of the human data come from studies on related conditions, such as bipolar disorder or neurodegenerative diseases, where uridine or TAU was used as an adjunct. The evidence is promising but not definitive, which is why a rating of 3 (moderate evidence) is appropriate. More robust, targeted clinical studies are needed to fully validate TAU’s role in mitochondrial support.
