Vitamin B1, also known as thiamine (often in the supplemental form thiamine mononitrate), is scientifically validated as essential for mitochondrial function. Thiamine acts as a critical cofactor for several mitochondrial enzymes involved in carbohydrate metabolism, most notably pyruvate dehydrogenase and α-ketoglutarate dehydrogenase, both of which are essential for the Krebs (citric acid) cycle. This cycle is fundamental for the production of ATP—the main energy currency of the cell—within the mitochondria. Deficiency in thiamine leads to impaired mitochondrial energy production, and this is clinically evident in disorders such as beriberi and Wernicke-Korsakoff syndrome, where neurological and cardiovascular symptoms can be traced to inadequate cellular energy production.

There is robust experimental and clinical evidence that thiamine supplementation can restore or support mitochondrial function in cases of deficiency. Studies in both animals and humans have shown that thiamine deficiency impairs mitochondrial respiration, and that supplementation reverses these effects. While supplementation beyond physiological needs in healthy individuals is not generally shown to enhance mitochondrial function, in states of deficiency or increased demand (such as certain metabolic disorders, chronic alcoholism, or specific inherited mitochondrial diseases), thiamine is a critical intervention. Thus, its use to support the mitochondrial system is based on well-established biochemical and clinical principles.
