Vitamin B2, also known as riboflavin, has strong scientific evidence supporting its role in mitochondrial function. Riboflavin serves as a precursor for the cofactors flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD), both of which are essential for the function of various mitochondrial enzymes, including those in the electron transport chain (complexes I and II). These enzymes are critical for oxidative phosphorylation, the process by which cells generate ATP, the main energy currency of the cell. Riboflavin deficiency impairs mitochondrial energy production, leading to symptoms such as fatigue and muscle weakness.

Clinical interventions with riboflavin have been shown to benefit certain mitochondrial disorders, such as multiple acyl-CoA dehydrogenase deficiency (MADD) and some forms of mitochondrial complex I deficiency, where riboflavin supplementation can improve biochemical and clinical outcomes. These effects are well-documented in both biochemical research and clinical case series. Therefore, riboflavin is scientifically validated as a critical nutrient for mitochondrial health and function.
