Guanidinoacetic acid (GAA) is a naturally occurring precursor to creatine, a compound critical for cellular energy metabolism, particularly in muscle tissue. Several studies have investigated GAA supplementation as a means to increase creatine levels, especially in individuals with impaired creatine biosynthesis or uptake. Some clinical and preclinical research has shown that GAA supplementation can raise intramuscular creatine stores, which may help support muscle energy metabolism and function. In populations with creatine deficiency syndromes or certain neuromuscular disorders, this pathway is of therapeutic interest.

However, direct evidence for GAA’s efficacy in improving muscle tone specifically (i.e., increasing or restoring normal muscle tension or firmness) is limited. Most studies focus on muscle strength, function, or mass rather than "muscle tone" per se. The evidence for GAA improving muscle tone in healthy individuals or in the context of common hypotonia is weak, with a few pilot studies indicating potential benefit in clinical populations with severe creatine deficiency. Overall, while GAA has a plausible biochemical rationale and some early clinical support for improving muscle function via creatine synthesis, high-quality, large-scale trials focused specifically on muscle tone are lacking. Therefore, the evidence supporting its use for muscle tone is modest and not yet robust.
