Ligandrol (also known as LGD-4033) is a selective androgen receptor modulator (SARM) that has been extensively researched for its effects on muscle mass and strength. Several clinical trials have demonstrated that Ligandrol can increase lean muscle mass and improve muscle strength in both healthy volunteers and populations at risk of muscle wasting, such as older adults. For example, a randomized, double-blind, placebo-controlled phase I trial published in "The Journal of Clinical Endocrinology & Metabolism" (2013) showed that healthy men who took LGD-4033 for 21 days experienced a dose-dependent increase in lean body mass without significant adverse effects. The mechanism involves selective activation of androgen receptors in muscle and bone tissue, which promotes protein synthesis and muscle growth while minimizing unwanted androgenic effects in other tissues.

However, while the evidence for its efficacy in increasing muscle mass and strength is robust, Ligandrol is not approved by major regulatory agencies (such as the FDA) for medical use and is classified as a prohibited substance by the World Anti-Doping Agency (WADA). Its safety profile for long-term use remains uncertain, and side effects such as suppression of natural testosterone production have been reported. Nevertheless, the scientific evidence for its role in improving muscle tone (by increasing muscle mass and strength) is strong, but its use should be limited to research settings and not as a general supplement or therapeutic agent until more safety data are available.
