Glycerol (also known as glycerin) is used in sports and exercise settings primarily for its role in promoting hyperhydration. Scientific studies have shown that glycerol, when ingested with water, can increase total body water by enhancing fluid retention. This effect is due to glycerol’s ability to increase the osmolarity of body fluids, thereby reducing urine output and helping maintain hydration status during prolonged physical activity. Improved hydration can support muscle function and endurance, as dehydration is known to impair muscular performance, increase fatigue, and potentially elevate the risk of muscle cramps or heat-related illnesses.

Several randomized controlled trials and systematic reviews have investigated glycerol supplementation in athletes. The consensus is that glycerol-induced hyperhydration can improve endurance performance in hot environments, although the benefit is less clear in cooler conditions or during short-duration exercise. Glycerol is not directly associated with muscle growth or strength, but its capacity to support optimal muscle function via improved hydration is supported by moderate scientific evidence.

Glycerol's use as a performance aid has been recognized by sports nutritionists, although it was temporarily placed on the World Anti-Doping Agency’s banned list due to potential masking effects on urine tests (it was removed from the list in 2018). In summary, the use of glycerol to support the muscle system is scientifically validated specifically through its indirect role in maintaining hydration and delaying fatigue during prolonged exercise.
