Nicotinamide Adenine Dinucleotide (NAD+) is a crucial coenzyme found in all living cells and is fundamental to energy metabolism, particularly in muscle tissue. Scientific research has shown that NAD+ plays a central role in mitochondrial function, facilitating oxidative phosphorylation and ATP production, which are essential processes for muscle contraction and endurance. As muscles require substantial amounts of energy for contraction and repair, maintaining optimal NAD+ levels is vital for muscle health and performance.

Emerging studies suggest that NAD+ levels decline with age, and this decline is associated with reduced mitochondrial function and muscle weakness. Preclinical research in animal models has demonstrated that supplementation with NAD+ precursors (such as nicotinamide riboside and nicotinamide mononucleotide) can improve muscle mitochondrial function, enhance exercise performance, and ameliorate age-related muscle degeneration. Early human trials have shown promising results regarding improved muscle energetics and endurance, but larger and more definitive clinical trials are still needed.

While the mechanistic rationale and preclinical evidence are strong, direct clinical proof in humans is currently limited. Therefore, the use of NAD+ or its precursors to support muscle function is scientifically plausible and supported by preliminary evidence, but the overall strength of evidence is moderate at this time.
