Vitamin B6 (pyridoxine) is involved in many aspects of nervous system function, particularly as a coenzyme in the synthesis of neurotransmitters such as serotonin, dopamine, and gamma-aminobutyric acid (GABA). This biochemical role has led to its investigation in conditions related to mood and mental fatigue. "Nervous exhaustion" is an older, non-specific term that can encompass symptoms of fatigue, depression, and anxiety. While Vitamin B6 deficiency is known to cause neurological symptoms such as irritability, depression, and confusion, evidence supporting its use specifically for "nervous exhaustion" in otherwise healthy, non-deficient individuals is limited. Some clinical studies have explored the effects of B6 supplementation on mood and cognitive function, with mixed results. Systematic reviews have found that B6 may have a small effect on reducing symptoms of premenstrual syndrome (PMS)-related mood disturbance, but robust evidence for benefits in general mental fatigue or burnout is lacking. In cases of diagnosed deficiency, supplementation is necessary and can resolve neuropsychiatric symptoms. However, for the general population experiencing "nervous exhaustion" not related to B6 deficiency, scientific support for supplementation is weak. Therefore, the use of Vitamin B6 for this indication is based on its biological plausibility and established role in neurotransmitter synthesis, but not on strong clinical trial evidence.
