Diosmetin is a flavonoid aglycone derived from diosmin, commonly found in citrus fruits. Recent preclinical studies have investigated its potential effects on osteoporosis. Laboratory (in vitro) research has shown that diosmetin can promote osteoblast differentiation and mineralization, which are critical processes for bone formation. Additionally, animal studies, particularly in ovariectomized rat models (which mimic postmenopausal osteoporosis), have demonstrated that diosmetin administration may help mitigate bone loss, improve bone microarchitecture, and increase bone mineral density. These effects are thought to occur via modulation of signaling pathways such as Wnt/β-catenin and inhibition of osteoclastogenesis, partially through anti-inflammatory and antioxidant activities. However, to date, there are no published human clinical trials evaluating diosmetin for osteoporosis prevention or treatment. The evidence remains limited to cell culture and animal models. While the mechanism of action is biologically plausible and early results are promising, the lack of high-quality human data limits the strength of the evidence supporting its use. Therefore, diosmetin’s use for osteoporosis is justified by preliminary scientific research but is not yet established as an effective or recommended therapy in humans.
