Casticin is a flavonoid compound found primarily in the fruit and leaves of Vitex agnus-castus, commonly known as chaste tree. The use of Vitex agnus-castus has a long tradition in herbal medicine, particularly in Europe, for supporting female reproductive health, including the management of menstrual disorders and support of ovarian function. The traditional use is based on historical texts and ethnobotanical reports, with the belief that compounds like casticin contribute to the plant’s purported ability to balance hormones, alleviate premenstrual syndrome (PMS), and regulate menstrual cycles.

Scientific studies on Vitex agnus-castus extracts have shown some promise in alleviating PMS and other menstrual issues, but the direct role of casticin itself in supporting ovarian function is less well established. Most research involves whole-plant extracts rather than isolated casticin, and the mechanisms of action are not fully understood. While casticin is considered one of the marker compounds for standardizing Vitex extracts, there is limited direct evidence that casticin alone has significant effects on the ovaries. Overall, support for its use is rooted in traditional practices, with only limited scientific validation for the whole plant and insufficient specific data for casticin as an isolated compound.
