Lupulinum is a historic herbal preparation derived from the glandular trichomes (lupulin) of the hops plant (Humulus lupulus). Traditionally, lupulin and hops have been used as mild sedatives and nervines, particularly in European and North American herbal medicine. The connection to the parasympathetic nervous system arises from this sedative action, as parasympathetic activity is associated with rest, digestion, and relaxation.

Historical texts and herbal compendia from the 19th and early 20th centuries describe lupulinum as being used to soothe nervous tension, promote sleep, and relieve anxiety. Its use was primarily empirical, based on observations of calming effects rather than on an understanding of neurophysiology. While hops contain compounds such as humulone, lupulone, and prenylflavonoids, which have demonstrated mild sedative and anxiolytic properties in animal studies, there is limited direct scientific validation specifically for lupulinum in relation to the parasympathetic nervous system.

Modern research into hops as a whole has suggested possible activity on GABAergic systems, which could theoretically enhance parasympathetic tone, but clinical evidence remains limited and does not specifically address lupulinum or direct effects on the parasympathetic nervous system. Thus, its use for supporting this body system is primarily justified by traditional herbal practices rather than robust scientific evidence.
