β-Nicotinamide mononucleotide (NMN) is a precursor to nicotinamide adenine dinucleotide (NAD+), a key molecule involved in cellular energy metabolism and mitochondrial function. Scientific interest in NMN for Parkinson's Disease (PD) arises from research showing that NAD+ levels decline with age and neurodegenerative conditions, possibly contributing to neuronal dysfunction and death. Preclinical studies in animal models of PD have demonstrated that boosting NAD+ levels, either via NMN supplementation or other NAD+ precursors, can improve mitochondrial function, reduce neuroinflammation, and protect dopaminergic neurons from degeneration. For example, some mouse studies suggest that NMN administration ameliorates motor deficits and preserves dopaminergic neurons in toxin-induced models of PD. Furthermore, NMN may enhance the activity of sirtuins and other NAD+-dependent enzymes, which are implicated in neuroprotection and cellular repair mechanisms.

However, direct clinical evidence in humans with PD is extremely limited. As of early 2024, there are no published large-scale clinical trials demonstrating that NMN supplementation provides significant benefit for PD patients. Most supporting evidence remains at the preclinical stage, and the translation of these findings to human disease is unproven. Thus, while the mechanistic rationale is supported by laboratory studies and early animal data, robust scientific validation in humans is lacking. The overall evidence for NMN's use in PD is therefore preliminary, justifying a rating of 2/5.
