Synopsis

Source of validity: Scientific
Rating (out of 5): 2

Uridine monophosphate (UMP) has some scientific evidence supporting its use in the management of peripheral neuropathy, though the overall quality and quantity of evidence is limited and generally rated as low to moderate. UMP is a nucleotide that plays a role in neuronal membrane synthesis and repair. The rationale for its use in neuropathy is based on its involvement in promoting nerve regeneration and enhancing the synthesis of phospholipids necessary for nerve cell membranes.

Several clinical studies, often conducted in combination with other neurotrophic factors such as vitamin B12 and folic acid, have reported improvements in symptoms of peripheral neuropathy, particularly in diabetic or chemotherapy-induced neuropathy. A few randomized controlled trials and observational studies have suggested that oral supplementation with UMP (sometimes combined with cytidine monophosphate and vitamin B12) may improve nerve conduction velocity, reduce pain, and enhance overall nerve function in affected individuals. However, most of these studies are small, industry-sponsored, and sometimes lack robust blinding or placebo controls.

Systematic reviews have concluded that while there is a plausible biological rationale and some clinical evidence, the data are not strong enough to firmly establish UMP as an effective treatment for peripheral neuropathy on its own. Larger, high-quality, independent trials are needed to confirm its efficacy and safety. Thus, its use is considered to have some scientific basis, but the evidence is relatively weak at present.
