MK-677 (Ibutamoren) is a non-peptide growth hormone secretagogue that stimulates the pituitary gland to increase secretion of growth hormone (GH) and insulin-like growth factor 1 (IGF-1). Its mechanism of action is well-established: MK-677 mimics the action of ghrelin, binding to the ghrelin receptor (GHSR) in the brain, which in turn stimulates the pituitary to release GH. Multiple clinical studies have demonstrated that MK-677 increases GH and IGF-1 levels in both healthy individuals and those with GH deficiencies. For example, a 1998 study in elderly subjects showed that daily oral administration of MK-677 significantly increased GH and IGF-1 without serious adverse effects.
