Agnuside is one of the primary iridoid glycosides found in Vitex agnus-castus (chaste tree), a plant traditionally used for women’s hormonal health, particularly for premenstrual syndrome (PMS) symptoms. The use of Vitex for PMS, specifically PMS Type A (characterized by anxiety, irritability, and mood swings), is supported by both historical use and modern clinical research. Scientific evidence indicates that the efficacy of Vitex is likely due to its dopaminergic effects, which can reduce prolactin secretion and help rebalance the luteal phase of the menstrual cycle. Agnuside is considered a key active constituent responsible for these effects, though it is often studied as part of the whole extract rather than in isolation. Several randomized, placebo-controlled trials and meta-analyses have shown that Vitex extracts are more effective than placebo for relieving PMS symptoms, including mood-related issues. The 2017 Cochrane review concluded that Vitex may be more effective than placebo, though the quality of evidence was moderate and further research is needed. While most evidence is for the whole extract, agnuside is widely cited in phytochemical analyses as a marker of extract quality and potential bioactivity.
