Acid proteases are enzymes that function optimally at low pH, such as those found in the stomach (e.g., pepsin). They are used in supplements to support protein digestion, especially in individuals with hypochlorhydria (low stomach acid), atrophic gastritis, or age-related decline in digestive function. Scientific evidence indicates that proteolytic enzymes, including acid proteases, can enhance protein breakdown in the stomach, improving amino acid absorption.

Several studies have shown that supplemental proteases can aid in the digestion of dietary proteins, particularly in people with compromised gastric function. For instance, research has demonstrated that protease supplementation increases the rate and extent of protein hydrolysis in vitro and can enhance nitrogen absorption in vivo. Clinical trials are somewhat limited but suggest benefits in individuals with pancreatic insufficiency or after gastric surgery, where endogenous enzyme production is reduced. Additionally, acid proteases are widely used in food processing to pre-digest proteins, indirectly supporting the rationale for their use in supplements.

However, for the general population with normal digestive function, the incremental benefit of acid protease supplementation is less clear, and most evidence is extrapolated from specific populations or laboratory studies.
