Andrographolide is a diterpenoid compound extracted from Andrographis paniculata, a plant used in traditional Asian medicine. There is emerging scientific interest in its potential application for psoriasis, a chronic autoimmune skin disorder characterized by inflammation and hyperproliferation of keratinocytes. Several preclinical studies have demonstrated that andrographolide possesses anti-inflammatory, immunomodulatory, and antioxidant properties, which are relevant to the pathogenesis of psoriasis. For instance, animal model studies have shown that topical or systemic administration of andrographolide can reduce inflammatory cytokine levels such as TNF-α, IL-17, and IL-23, all of which are implicated in psoriatic pathology. Additionally, andrographolide has been observed to inhibit the activation of NF-κB, a key regulator of inflammation in psoriasis. However, clinical evidence in humans remains limited. A few small-scale clinical trials and case reports have suggested some benefit in reducing the severity and symptoms of psoriasis, but these studies often lack rigorous design or have small sample sizes. No large, well-controlled randomized clinical trials have yet confirmed its efficacy for this indication. In summary, while there is a scientific rationale and some supportive preclinical evidence for the use of andrographolide in psoriasis, the clinical evidence is currently insufficient to strongly recommend its use. More robust human studies are required to determine its safety and efficacy for this condition.
