Hyperoside is a flavonoid glycoside found in various medicinal plants, such as Hypericum perforatum (St. John’s Wort), Crataegus species (hawthorn), and others. Recent preclinical studies have explored its potential anti-inflammatory and immunomodulatory effects, which are relevant to the pathogenesis of Rheumatoid Arthritis (RA). Several in vitro and animal studies suggest that hyperoside can inhibit the production of pro-inflammatory cytokines (such as TNF-α, IL-1β, and IL-6), suppress the activation of NF-κB signaling pathways, and reduce oxidative stress in models of inflammatory diseases, including experimental arthritis in rodents.

For example, a 2020 study in Frontiers in Pharmacology demonstrated that hyperoside attenuated collagen-induced arthritis in mice by reducing joint inflammation and cartilage destruction, likely through suppression of inflammatory mediators and modulation of immune cell infiltration. Other laboratory studies have reported similar findings, indicating reduced joint swelling and improved histopathological scores in hyperoside-treated animals.

However, there is currently a lack of clinical trials in humans evaluating hyperoside’s efficacy or safety in the treatment of RA. Most supporting evidence is limited to experimental models, and the translation of these results to human disease remains uncertain. Thus, while there is some scientific rationale and preclinical support, clinical validation is lacking, and the overall evidence remains preliminary.
