Nobiletin is a polymethoxylated flavone predominantly found in citrus peels. Recent preclinical studies have investigated its potential anti-inflammatory and immunomodulatory effects relevant to rheumatoid arthritis (RA). In animal models of RA, such as collagen-induced arthritis in mice, nobiletin has demonstrated a capacity to reduce joint swelling, decrease pro-inflammatory cytokines (e.g., TNF-α, IL-1β, IL-6), and mitigate cartilage destruction. The proposed mechanisms include inhibition of the NF-κB signaling pathway and downregulation of matrix metalloproteinases, both implicated in RA pathogenesis.

However, the evidence supporting nobiletin’s use in RA is currently limited to in vitro studies and animal models. No published human clinical trials have directly assessed its efficacy or safety in RA patients. Additionally, while some traditional medicine texts mention citrus peels for general inflammation, the specific use of nobiletin for RA is not documented in traditional medical literature.

In summary, nobiletin shows promising anti-inflammatory effects in preclinical models relevant to RA, but there is a lack of human clinical data. Thus, while there is a scientific rationale for further investigation, current evidence supporting its use for RA is preliminary, and it should not be considered a validated treatment at this time.
