Synopsis of Evidence Supporting Polydatin for Rheumatoid Arthritis

Polydatin, a natural glycoside of resveratrol found in Polygonum cuspidatum and other plants, has attracted scientific interest for its anti-inflammatory and antioxidant properties. Several preclinical studies have investigated its effects in models of rheumatoid arthritis (RA). For example, animal studies have shown that polydatin administration can reduce joint inflammation, cartilage destruction, and expression of pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6 in collagen-induced arthritis models in mice. Mechanistically, it is believed to exert these effects through inhibition of the NF-κB signaling pathway, modulation of oxidative stress, and suppression of inflammatory mediators.

However, the evidence is currently limited to in vitro experiments and animal models; there are no published clinical trials evaluating the efficacy or safety of polydatin in human patients with RA. While these findings are promising and suggest a potential therapeutic role, the absence of human data means that its use cannot yet be recommended as standard therapy. Most existing research has been published in the last decade and is exploratory in nature. Therefore, the evidence supporting the use of polydatin in RA can be considered preliminary (rated 2 out of 5), pending further clinical research.
