Glycyrrhizin, a major active component of licorice root (Glycyrrhiza glabra), has been studied for its anti-inflammatory and immunomodulatory properties, which are relevant in the context of rhinitis, particularly allergic rhinitis. Some preclinical studies and limited clinical research suggest that glycyrrhizin can inhibit the release of histamine and pro-inflammatory cytokines, such as IL-4 and IL-5, from mast cells and other immune cells involved in the pathogenesis of allergic rhinitis. In animal models, glycyrrhizin has demonstrated the ability to suppress nasal symptoms and reduce eosinophilic infiltration in nasal tissues. Additionally, glycyrrhizin may modulate the Th1/Th2 balance, favoring an anti-allergic immune response. A few small human studies and clinical trials have investigated the use of licorice extracts containing glycyrrhizin in patients with allergic rhinitis, reporting improvements in nasal symptoms and reductions in inflammatory markers. However, these studies are generally limited by small sample sizes, short durations, and lack of rigorous controls. There is also a paucity of large-scale, high-quality randomized controlled trials directly assessing purified glycyrrhizin for rhinitis. In summary, while there is scientific rationale and preliminary evidence supporting glycyrrhizin’s use in rhinitis, the overall level of clinical evidence is low to moderate, and further robust studies are needed to confirm its efficacy and safety for this indication.
