5-Methoxytryptamine (5-MT), also known as mexamine, is an endogenous tryptamine derivative structurally related to serotonin and melatonin. Scientific interest in 5-MT centers on its role as a serotonin receptor agonist, particularly at 5-HT1 and 5-HT2 subtypes. Preclinical studies have shown that 5-MT can cross the blood-brain barrier and interact with serotonin receptors, influencing physiological processes associated with the serotonergic system, such as mood, circadian rhythms, and neuroendocrine regulation. However, direct human clinical evidence supporting the use of 5-MT as a supplement to support the serotonin system is limited. Animal studies have demonstrated that 5-MT administration may affect sleep-wake cycles and modulate neurotransmitter activity, suggesting a potential for interaction with the serotonin system. Despite these findings, 5-MT is not widely used in clinical practice or dietary supplements for this purpose, and there are few (if any) robust human trials confirming efficacy or safety. Most of the available evidence comes from basic neuroscience research rather than applied clinical use. In summary, while there is a scientific rationale based on mechanistic data and animal studies for the involvement of 5-MT in the serotonin system, the evidence is limited and mostly preclinical. Therefore, its use to support the serotonin system is based more on experimental findings than established clinical validation.
