Hyperforin is a key active compound found in the plant Hypericum perforatum, commonly known as St. John's Wort. The use of St. John's Wort for mood support and emotional health is well established both traditionally and in contemporary clinical settings. The scientific interest in hyperforin centers around its mechanism of action regarding neurotransmitter modulation, particularly its effects on the serotonin system.

Multiple in vitro and animal studies have shown that hyperforin inhibits the reuptake of several neurotransmitters, including serotonin, dopamine, noradrenaline, GABA, and glutamate. This broad reuptake inhibition is primarily attributed to hyperforin, rather than to other constituents of St. John's Wort. This mechanism is similar to that of conventional selective serotonin reuptake inhibitors (SSRIs), which are widely used antidepressant medications.

Clinical trials and meta-analyses have demonstrated that St. John's Wort extracts, standardized to hyperforin content, are more effective than placebo and similarly effective to standard antidepressants in cases of mild to moderate depression. However, most studies use whole plant extracts rather than isolated hyperforin, so direct data on hyperforin alone are limited. Still, the evidence strongly suggests that hyperforin plays a significant role in the observed serotonergic effects.

In summary, the use of hyperforin to support the serotonin system is primarily justified by scientific evidence, with a strong mechanistic basis and supportive clinical trial data involving St. John's Wort extracts.
