Vitamin B6 (pyridoxine and related vitamers) plays a well-established role in supporting the serotonin system through its function as a coenzyme in neurotransmitter synthesis. Specifically, Vitamin B6 is required for the activity of aromatic L-amino acid decarboxylase, the enzyme that converts 5-hydroxytryptophan (5-HTP) into serotonin. Deficiency in Vitamin B6 can impair serotonin production, potentially contributing to mood disorders such as depression. Several clinical and preclinical studies have demonstrated that supplementation with Vitamin B6 can raise serotonin levels or improve mood in certain populations, especially when baseline levels are low.

A 2004 review in the American Journal of Clinical Nutrition noted the biochemical necessity of B6 for serotonin synthesis and referenced studies where B6 supplementation improved depressive symptoms. More recent randomized controlled trials have investigated Vitamin B6 supplementation for mood support, with some finding moderate benefits, though effects are modest compared to prescription antidepressants.

While Vitamin B6 is not a standalone treatment for serotonin-related disorders, its essential cofactor role in serotonin biosynthesis and its modest benefit in mood support are supported by biochemical, clinical, and epidemiological evidence.
