L-Glutathione is a tripeptide composed of glutamine, cysteine, and glycine, and is well known as a major endogenous antioxidant in the human body. Its use in general skin care, particularly for skin lightening and anti-aging, has become popular in supplements and topical formulations. The rationale for its use is rooted in its ability to neutralize free radicals and reactive oxygen species, theoretically protecting skin cells from oxidative stress and reducing signs of aging. Several in vitro and small clinical studies suggest that oral and topical glutathione may have a modest effect on skin pigmentation, primarily through inhibition of melanin synthesis (melanogenesis). For example, a few randomized controlled trials have reported mild skin lightening effects after 4-12 weeks of oral supplementation, though the magnitude of effect is typically small and not consistent across all studies.

However, the overall quality and quantity of clinical evidence remains limited. Many studies have small sample sizes, short durations, and are sometimes industry-sponsored. There are also concerns about the bioavailability of oral glutathione and whether it reaches the skin in sufficient concentrations to exert meaningful effects. While L-glutathione is safe for most people, robust evidence supporting its general use for improving skin health, aside from potential mild lightening, is lacking. Therefore, while there is some scientific rationale and preliminary data, the evidence supporting its use in general skin care is weak to moderate at best.
