Lipase is an enzyme that plays a crucial role in the digestion and absorption of dietary fats in the small intestine. Scientific evidence strongly supports its physiological function: lipase, primarily secreted by the pancreas, hydrolyzes triglycerides into free fatty acids and monoglycerides, which can then be absorbed by the intestinal mucosa. In cases of exocrine pancreatic insufficiency (EPI)—such as chronic pancreatitis, cystic fibrosis, or pancreatic cancer—patients cannot produce enough pancreatic enzymes, including lipase. This deficiency leads to fat malabsorption, steatorrhea, and nutritional deficiencies. Supplementing with exogenous lipase as part of pancreatic enzyme replacement therapy (PERT) is a standard, well-supported medical intervention to restore fat digestion and absorption in these individuals. Numerous clinical studies and systematic reviews demonstrate that PERT, which includes lipase, significantly improves nutrient absorption and gastrointestinal symptoms in people with EPI. However, outside of diagnosed enzyme deficiencies, the benefit of supplemental lipase for the general population or for nonspecific digestive complaints is not well-supported by high-quality evidence. In summary, there is robust scientific validation for the use of lipase to support the small intestine's digestive function in individuals with pancreatic enzyme insufficiency.
