Nicotinamide (also known as niacinamide) is a form of vitamin B3 that plays a crucial role in cellular metabolism and energy production as a precursor of NAD+ (nicotinamide adenine dinucleotide), which is essential for redox reactions in the brain. There is some scientific evidence to suggest that nicotinamide may influence neurotransmitter systems indirectly by supporting neuronal health and function. For instance, studies have shown that NAD+ depletion may impair neuronal survival and can be associated with neurodegenerative processes, while its replenishment (via nicotinamide supplementation) may confer neuroprotective effects. Additionally, some animal and cell studies indicate that nicotinamide can modulate the activity of enzymes involved in neurotransmitter synthesis and degradation, such as monoamine oxidase, and may affect levels of neurotransmitters like dopamine and serotonin. However, high-quality human trials directly linking nicotinamide supplementation to improved neurotransmitter function or neuropsychiatric outcomes are limited. The bulk of the evidence is preclinical or based on mechanistic reasoning rather than robust clinical results.
