Griffonia simplicifolia is a West African plant whose seeds are a natural source of 5-hydroxytryptophan (5-HTP), a direct precursor to the neurotransmitter serotonin. Serotonin is implicated in mood regulation, anxiety, and stress response. Several small clinical studies and animal experiments have investigated the effects of 5-HTP supplementation on mood and stress-related symptoms. Some evidence suggests that oral 5-HTP can improve mood and reduce symptoms associated with stress, anxiety, and depression, likely due to increased serotonin synthesis in the brain. However, the majority of these studies use isolated 5-HTP rather than Griffonia simplicifolia seed extract specifically, and sample sizes are often small.

There is limited direct clinical evidence on Griffonia simplicifolia itself for stress, but its use is scientifically rationalized by its 5-HTP content. The European Food Safety Authority (EFSA) and other regulatory bodies note insufficient robust evidence for health claims related to stress or mood. Overall, while there is a plausible mechanism and some supportive data for 5-HTP (and by extension Griffonia simplicifolia) in stress reduction, the quality and quantity of direct clinical evidence remain limited, justifying a modest evidence rating.
